Son of Samuel Wyckoff, John moved with his parents from Kentucky to Sangamon County, Illinois, in October 1822, settling in what became Chatham Township. In 1835, he purchased approximately 42 acres of public land in the area. Wyckoff married three times. In 1838, he married Sarah Shelton; they had no children. He wed Amanda Jacobs in 1843; the couple had one child. In 1850, he married Margarite Jane Foster.
