Busher was educated and completed an apprenticeship as a leather finisher in England before immigrating to the United States in 1833. In 1839, he settled in Springfield, Illinois, where he established John Busher & Co., manufacturer and dealer in harness and saddlery; he also later operated a tannery and a brewery in the city. In 1842, he married Emma Everson, with whom he had four children before her death in 1849. In 1852, he re-married to Emily B. Wyatt, with whom he had three children. In 1860, Busher owned real estate worth $10,000 and personal property worth $3,000. Politically, he was a Democrat.
