In 1817, Harbour moved with his brother Levi from Christian County, Kentucky, to Madison County, Illinois. By 1819, Harbour had settled in Sangamon County, Illinois, west of Chatham, where he would reside for the rest of his life. In 1823, he married Elizabeth Briscoe, with whom he had one child before her death in 1824. Two years later, Samuel re-married to Elizabeth Lindley, with whom he had nine children. Harbour was a farmer; in 1850, he owned $2,400 worth of real estate.
