Nuckolls moved from Virginia to Illinois in 1818 with his parents and siblings. In 1820, they settled in Sangamon County. Charles married Mary Wilson in 1832, and together they had eight children. Nuckolls was a farmer and builder, and later had a leather-tanning business in Springfield. Nuckolls studied medicine late in life, graduating from McDowell College in St. Louis in 1856. He practiced medicine for several years before his death.
