Frye practiced medicine in Peoria, Illinois for fifty-three years. In 1849, he organized the Peoria Medical Association and served as its first president. He was also a member of the State Medical Society of Illinois. Frye supported the Whig Party. In 1860, Frye owned $26,000 in real estate and had a personal estate of $1,200. Frye and his wife, Eliza, had one son and three daughters.
