Gildersleeve grew up near Hempstead, New York. He briefly studied law there, and on August 23, 1828, he married Mary Ann Eckford Rhodes. The couple and one child and settled in Illinois in 1836. From 1845-49, Gildersleeve was a clerk of the McLean County Circuit Court. After Mary died, James married Elizabeth S. Conkling on October 14, 1847. In 1850, Gildersleeve was a farmer in McLean County, Illinois, where he owned $4,000 in real property.
