William Pope was a merchant, clerk, and son of Nathaniel Pope. Sometime prior to the Mexican War he lived in Alton, Illinois, where he co-owned a drugstore with his brother-in-law, Thomas M. Hope. Pope lived in Sangamon County, Illinois, from at least 1845 to his death in 1855. In 1850, he was employed as clerk of the federal district court in Springfield, where he lived with his wife and children.
